Molecular Identification, Prevelance and Antimicrobial Susceptibility Profile of Cronobacter spp. Cultivated on a Chromogenic Medium in Libya.

Background: Cronobacter sakazakii is associated with illness in infants from contaminated powdered infant formula (PIF) and it is frequently recovered from PIF factory environment. Limited information is available on contamination of other food such as dairy and meat products in Libya. Methods and Findings: A total of 261 samples of milk, dairy products and coarse ground meat products were collected from different localities in Libya. Samples were examined for Cronobacter spp. with an adapted ISO /DTS 22964 cultural protocol using HiChrome™ Enterobacter sakazakii modified agar coupled with 16S rDNA partial sequencing to identify the organism. The identified isolates were biochemically characterized and tested for their ability to produce yellow pigment. Out of the 261 analyzed samples, only two beef burgers, one fermented milk “Laban”, one she-camel’s milk, two raw cow’s milk, two cereal baby food, one Maassora cheese and one ready to feed baby milk were contaminated with Cronobacter spp. at a total rate of 3.8%. Accuracy of HiChrome Ent. sakazakii modified agar reach 100% as all of blue-green presumptive colonies were confirmed Cronobacter spp. while other colorless, greenish or with blue center colonies which competed growth with Cronobacter spp. were predominantly Escherichia coli followed by Klebsiella spp. and to less extent Pseudomonas luteola, Citrobacter freundii and Acinetobacter baumanii. Moreover, the isolated strains of Cronobacter were resistant to Amoxicillin, Erythromycin, Vancomycin and Streptomycin, and sensitive to Doxycycline, Enrofloxacin and Gentamycin. Conclusion: This study documents for the first time the occurrence of Cronobacter spp. in beef burger, raw cow’s mi …

T lymphocyte subpopulations and B lymphocyte cellsin caecum and spleen of chicks infected with Salmonella enteritidis

The effect of low and high doses of Salmonella enteritidis PT4 (SE) on immunocompetent cells in caecum and spleen of one-day-old chicks was investigated. Subsets of T lymphocytes positive for CD3, CD4, CD8 and B lymphocytes (Bu1b-positive cells) were counted in the caecum after immunohistochemical staining and the relative percentage of these cells in the spleen was analysed using a FACScan cytometer on days 7, 10, 14, 21, and 27 post-inoculation (pi). In the low dose group, the number of CD3+ and CD4+ cells in the caecum had significantly increased at day 10 pi. Both CD8+ and Bu1b+ cells were significantly higher on day 14 pi in this group. In the high-dose group, the number of CD4+ cells had significantly increased at day 7 pi. CD3+, CD8+, and Bu1b+ cells showed prolonged proliferation at days 7 up to 21 pi. Splenic lymphocytes demonstrated significant changes only in the high dose group. The percentage of splenic CD4+ cells was decreased at day 7 pi. A decrease in CD3+ and CD8+ cells was found at day 14 pi in this group.

Seroprevalence and potential risk factors associated with Toxoplasma gondii infection in women from Tripoli, Libya

Aims: To determine the seroprevalence of Toxoplasma gondii (T. gondii) in women from Aljalla Maternity and Gynaecology Hospital (AMGH) and to evaluate the association between the infection and potential risk factors. Methods: A cross-sectional study was conducted at AMGH in Tripoli during the year 2012. Data on potential risk factors were collected by a structured questionnaire and results of the test. The output data of questionnaire were used to assess potential risk factors for positive outcome. All sera (n = 500) was screened against ToxoG, by using the Vidas machine automated enzyme-linked florescent immunoassay. The association between the outcome variables and its potential risk factors were screened in a multivariate analysis. A p-value < 0.05 was considered to be significant. Results: The overall seroprevalence was estimated to be 50.8% with 95% confidence interval (CI) (46.42%–55.18%). The results revealed that T. gondii infection is highly prevalent, and eating habits (consuming raw meat and/or raw vegetables or fruits) were highly significant associated in multivariable analysis. Expectedly, a significantly [odds ratio (OR):1.712; p =
